Synthesis of steroidal anti-tumor agents: Cephalostatins and saponin OSW-1

Abstract

Syntheses of eleven natural and unnatural cepholastatins are described herein. Five related projects are included in this thesis: (1) improvement of the syntheses of both halves of cephalostatin 7 (1.7); (2) Development of an efficient protocol for unsymmetrical pyrazine synthesis; (3) Completion of the first total synthesis of cephalostatin 1 (1.1) and preparation of other ten cephalostatin analogs employing the new pyrazine coupling method; (4) Establishment of a concise synthetic route leading to the aglycone of saponin OSW-1 (1.44); (5) Analysis of the SAR data obtained from cytotoxicity testing on the synthetic cephalostatins.
