Rapid confirmation of protein structure

Abstract

Most of the current techniques in protein structure analysis are suitable for protein structure elucidation. With the development of biotechnology, there is a need in developing techniques for protein structure confirmation which include the confirmation of product structure and the confirmation of the absence of product variants. Frontal Immunoaffinity Chromatography (FIC) has been developed for this purpose. FIC has been shown to be able to rapidly confirm protein structure, discriminate protein variants and determine antibody titer. Electrospray Mass Spectrometry was explored to be able to confirm protein conformation rapidly. Other techniques such as high performance liquid chromatography and capillary electrophoresis in different separation modes have also been shown their promise in rapid protein variants discrimination. The coupling technique between HPLC, HPCE, immunoassay and mass spectrometry will offer the most powerful tool in rapid protein structure confirmation.
