Design and synthesis of memapsin 2 inhibitors for treatment of Alzheimer's disease
Abstract
Alzheimer's disease (AD) is a neurodegenerative disorder affecting millions of people worldwide. My thesis work involved the design and synthesis of memapsin 2 inhibitors. Memapsin 2 is one of the transmembrane proteins that cleaves the amyloid precursor protein (APP) to form amyloid- β (Aβ), leading to the formation of insoluble amyloid plaques that are predominantly present in patients affected with AD. Three series of inhibitors were synthesized based on previous investigation carried out by Ghosh and co-workers; targeting the S2, S3, and S1' domains of the aspartic protease to achieve optimum potency.
