LC-MS/MS based metabolic profiling of breast cancer cells
Abstract
Metabolomics is a readily growing scientific area which focuses on the analysis of small molecule metabolites for various applications including early cancer detection. Mass spectrometry stands out among a number of metabolomics methodologies because of its high sensitivity and versatility. Liquid chromatography triple quadrupole mass spectrometry (TQMS), with excellent linear dynamic range and quantitation ability, is an ideal tool for targeted metabolomics research. This thesis describes the development of a TQMS based targeted LC-MS/MS method, as well as its application to the quantitative analysis of cancer cell samples. The instrument and targeted assay that incorporates 20 metabolites (19 of which with good linearity) in standard solutions in positive and negative mode are described. The analysis of cancer cell samples along with the results indicating potential biomarkers for the differentiation between cancer and normal breast cells are presented.
